Flow Cytometry Analysis and Cell Sorting Facility

The MoFlo Flow Cytometry Laboratory in the Department of Microbiology, Immunology and Pathology is a multi-user facility located in the CSU Pathology building room 101B.

Instrumentation
MoFlo (Dako Colorado, Inc.) Flow Cytometer and High Speed Cell Sorter.

  1. Solid state iCyt 488nm (blue) laser
  2. 635nm (red) diode laser
  3. Coherent Radius 405nm (violet) laser
  4. Capable of measuring up to 9 fluorescent parameters in addition to forward and side light scatter.
  5. Sort as many as 4 sub-populations of cells at one time, rather than the standard 2 fractions.
  6. Sort single cells (or any specified number) into 96 well plates using the automated Cyclone attachment. Other sizes of plates and trays can also be used, and microscope slides.

Due to the complexity of this instrument, all analysis and cell sorting experiments need to be scheduled with the trained operator. The purity and yield of the sort can be maximized according to client needs.

Services Available
Many possibilities exist for flow cytometric analysis or individual cell sorting, including, but not limited to:

  1. Immunofluorescent measurements (surface and intracellular markers, single and multi-parameter) for antigen and antibody distributions (up to 9 colors with 3 lasers).
  2. Determination of cell viability using Propidium Iodide, or PI with Sybr 14, and Dead/Live cell gating.
  3. Expression of Green Fluorescence Protein (GFP), or ds-Red.
  4. Detection of apoptosis with TUNEL or Annexin-V.Cytomerty Analysis
  5. Multicolor identification or sorting of bacteria, parasites, neurons, sperm or RBC’s.
  6. Quantitative analysis of the number of molecules of a particular antigen found on the surface of a cell (MESF).
  7. Cell division studies using antibodies to bromodeoxyuridine (BrdU).
  8. DNA and cell cycle analysis, DNA Index for aneuploidy.
  9. Mutant isolation
  10. Plant cell studies
  11. Virus infected cells
  12. Sterile sorting into multi-well plates, trays or onto individual microscope slides with automated cloning device.
  13. One, two, three or four way sterile tube sorting.

Charges and Fees
The rates for service for CSU affiliated labs:

  1. Analysis is $80 an hour, with a ½ hour set up charge ($40 for set up).
  2. Sorting is $100 an hour, with a 1 hour set up charge ($100 for set up).
  3. Please be aware that if you cancel your appointment at the last minute, you are still charged for the set up time. Sorting/analysis charges start at the time of your appointment, not when you arrive.
  4. If you need to cancel your appointment, please do so as early as possible. If you are running late for your appointment, please notify Leslie so she can see if she can arrange for a later starting time for people scheduled after you.

Please have the following form completed for billing information the first time you come to the MoFlo lab. This form does not have to be filled out each time, only on fiscal year change or if your fund number changes.
